Resumen

In this study we applied the faecal egg count reduction test (FECRT), ITS2 and β-tubulin isotype 1 (β-tub-1) deep amplicon sequencing to assess, for the first time, the patterns of benzimidazole (BZD) resistance in gastrointestinal nematode (GIN) species infecting cattle from large-scale farms in Argentina subjected to different anthelmintic treatment strategies.

British breed cattle and their crossbreeds, representative of the beef production systems in Buenos Aires province, Argentina, were used for fecal sampling over a period of approximately two years across six commercial farms operating under a grazing-based finishing system. The efficacy of BZDs and IVM in each commercial farm included in the project was previous to the feces sampling for isotype-1 b-tubulin analyses.In order to assess the molecular identification of GIN and determine the BZD resistance status of GIN parasitizing livestock, fecal samples from calves were collected between April 2020 and October 2021. The frequency of benzimidazole resistance alleles in parasitic nematodes communities was determined through deep amplicon sequencing of the isotype-1 b-tubulin and ITS2 genes, as previously described and detailed in Avramenko et al. (2019). The prepared pooled library was run on an Illumina Desktop Sequencer using a 500-cycle paired-end reagent kit (MiSeq Reagent Kit, v2, MS-103–2003) at a final concentration of 15 pM, with the addition of 25% PhiX control v3 (Illumina, FC-110-3001). The MiSeq was set to generate only FASTQ files.
